目的探讨雷帕霉素对内毒素性肝损伤小鼠肝组织Bcl-2表达的影响。方法健康雄性昆明小鼠36只,随机分为正常组(12)、模型组(12)和雷帕霉素组(12)。应用脂多糖10mg/kg腹腔注射小鼠诱导内毒素肝损伤模型。造模后6h取血,检测小鼠血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活性,免疫组织化学方法和Western blot方法检测各组小鼠肝组织Bcl-2的表达。结果模型组血清ALT、AST含量明显高于正常组(P<0.01);雷帕霉素组血清ALT、AST含量明显降于模型组(P<0.05)。与正常组相比,模型组小鼠肝组织Bcl-2表达显著降低(P<0.01);而与模型组相比,雷帕霉素组小鼠肝组织Bcl-2表达显著升高高于(P<0.01)。结论雷帕霉素能够上调内毒素性肝损伤小鼠肝组织Bcl-2的表达。
Objective To investigate the effect of rapamycin on the expression of Bcl-2 in the liver of endotoxin-induced liver injury in mice. Methods Thirty-six healthy male Kunming mice were randomly divided into normal group (12), model group (12) and rapamycin group (12). Lipopolysaccharide 10mg / kg intraperitoneal injection of endotoxin induced liver injury model in mice. The blood samples were collected 6 h after the model establishment. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity were measured. The expression of Bcl-2 in the liver of each group was detected by immunohistochemistry and Western blot. Results Serum levels of ALT and AST in model group were significantly higher than those in normal group (P <0.01). Serum levels of ALT and AST in rapamycin group were significantly lower than those in model group (P <0.05). Compared with the normal group, the expression of Bcl-2 in the model group was significantly lower than that in the normal group (P <0.01), while the expression of Bcl-2 in the rapamycin group was significantly higher than that of the model group P <0.01). Conclusion Rapamycin could up-regulate the expression of Bcl-2 in the liver of endotoxin-induced liver injury mice.
