目的:研究黄芪注射液中多糖的测定方法和情况。方法:采用高效凝胶渗透色谱法,测定了246批黄芪注射液样品。色谱柱为TSK-GEL G4000PWXL(7.8mm×300mm);流动相为0.02%叠氮化钠溶液;流速为0.5mL·min-1;柱温40℃;示差折光检测器。结果:246批样品中多糖的重均分子量(Mw)在7684~108846之间,分子量分布宽度(Mw/Mn,Mn为数均分子量)在0.89~2.71之间。结论:该方法可用于黄芪注射液中多糖分子量及其分布的测定;测定结果可为黄芪注射液质量标准的提高提供依据。
Objective: To study the method and situation of polysaccharide determination in Astragalus injection. Methods: High performance gel permeation chromatography was used to determine 246 samples of Astragalus injection. The chromatographic column was TSK-GEL G4000PWXL (7.8mm × 300mm); the mobile phase was 0.02% sodium azide solution; the flow rate was 0.5mL · min-1; the column temperature was 40 ℃; differential refractive index detector. Results: The weight average molecular weights (Mw) of polysaccharides in 246 samples ranged from 7684 to 108846, and the molecular weight distribution width (Mw / Mn, Mn was the number average molecular weight) ranged from 0.89 to 2.71. Conclusion: This method can be used for the determination of molecular weight and distribution of polysaccharides in Radix Astragali. The results can provide the basis for improving the quality standard of Radix Astragali.
