目的:观察食管鳞癌、癌旁及正常食管组织中基质金属蛋白酶3(MMP-3)、基质金属蛋白酶9(MMP-9)和基质金属蛋白酶抑制剂3(TIMP-3)的表达情况,分析它们与食管鳞癌临床病理特征的关系。方法:采用免疫组织化学方法检测89例食管鳞癌组织(食管鳞癌组)、30例食管鳞癌癌旁组织(癌旁组)及30例正常食管组织(食管正常组织组)中MMP-3、MMP-9和TIMP-3的表达,并结合食管鳞癌临床病理学资料进行分析。结果:MMP-3、MMP-9和TIMP-3的表达率食管鳞癌组明显高于癌旁组及食管正常组织组(P<0.05),在癌旁组及正常食管组织组之间表达率差异无统计学意义(P>0.05)。MMP-3、MMP-9在食管鳞癌中的表达与患者性别、年龄、病理分级无关(P>0.05),与浸润深度、淋巴结转移、TNM分期相关(P<0.05)。TIMP-3在食管鳞癌中的表达与患者性别、年龄、病理分级及浸润深度无关(P>0.05),与淋巴结转移、TNM分期有关(P<0.05);食管鳞癌中MMP-3的表达与TIMP-3的表达呈负相关(r=-0.383,P=0.000)。结论:临床上检测MMP-3、MMP-9及TIMP-3蛋白的表达有助于食管癌的早期诊断及判断预后。
OBJECTIVE: To observe the expression of matrix metalloproteinase 3 (MMP-3), matrix metalloproteinase 9 (MMP-9) and matrix metalloproteinase 3 (TIMP-3) in esophageal squamous cell carcinoma, paracancer and normal esophageal tissues, Relationship with clinicopathological features of esophageal squamous cell. Methods: Immunohistochemistry was used to detect the expression of MMP-3 in 89 cases of esophageal squamous cell carcinoma (ESCC), 30 cases of esophageal squamous cell carcinoma (paracancer) and 30 cases of normal esophageal tissue (esophageal normal tissue) , MMP-9 and TIMP-3, and combined with clinicopathological data of esophageal squamous cell carcinoma. Results: The positive rate of MMP-3, MMP-9 and TIMP-3 expression in esophageal squamous cell carcinoma was significantly higher than that in paracancerous tissues and normal esophageal tissues (P <0.05) The difference was not statistically significant (P> 0.05). The expression of MMP-3 and MMP-9 in esophageal squamous cell carcinoma had no correlation with gender, age and pathological grade (P> 0.05), but was correlated with the depth of invasion, lymph node metastasis and TNM staging (P <0.05). The expression of TIMP-3 in esophageal squamous cell carcinoma was not related to the gender, age, pathological grade and depth of invasion (P> 0.05), but also to the lymph node metastasis and TNM stage (P <0.05) Negatively correlated with the expression of TIMP-3 (r = -0.383, P = 0.000). Conclusion: The clinical detection of MMP-3, MMP-9 and TIMP-3 protein expression is helpful for the early diagnosis of esophageal cancer and prognosis.
