目的 探索 p16基因在脑胶质瘤发生发展过程中的作用及 p16基因在脑胶质瘤临床诊断、治疗中的应用前景。方法 采用脂质体转染的方法 ,将外源野生型 p16基因导入胶质瘤细胞株 U2 5 1、C6 ,筛选阳性克隆。同时以空载体质粒 p CDNA3为对照 ,免疫组化检测 p16基因表达 ,用MTT法测定瘤细胞生长曲线及对转染的瘤细胞在裸鼠体内形成瘤块的变化进行分析。结果 转染 p16基因的 U2 5 1、C6细胞有外源 p16基因的整合及表达 ,克隆形成率减少 ,生长速度明显减慢 ,瘤细胞在裸鼠中形成瘤块体积显著降低。结论 导入外源野生型 p16基因可抑制胶质瘤细胞恶性增殖。
Objective To explore the role of p16 gene in the development of glioma and the potential of p16 gene in the clinical diagnosis and treatment of gliomas. Methods The wild-type p16 gene was transfected into glioma cell line U2 5 1, C6 by lipofection. The positive clones were screened. At the same time, the empty plasmid pCDNA3 was used as control, the expression of p16 gene was detected by immunohistochemistry, the growth curve of tumor cells was detected by MTT assay, and the change of tumor cells in nude mice was analyzed. Results The integration and expression of exogenous p16 gene were found in U251 and C6 cells transfected with p16 gene. The rate of clone formation and the growth rate were significantly decreased. The volume of tumor cells in nude mice was significantly reduced. Conclusion The introduction of wild-type p16 gene can inhibit the malignant proliferation of glioma cells.
