目的探讨人甲状旁腺激素1-34[human parathyroid hormone(1-34),hPTH1-34]对去卵巢大鼠骨质疏松症的治疗作用。方法 分别予去卵巢6周的Wister大鼠hPTH1-34(40μg·kg-1·d-1,sc每周5次)或生理盐水干预8周,假手术大鼠不予治疗。留取 腰椎1-6(L1-6)、右侧股骨及血、尿标本分别进行骨密度、骨生物力学和生化指标的测定。结果 hPTH1-34组L1-6、股骨远端和股 骨干骨密度值明显高于去卵巢对照组,且L1-6和股骨干的骨密度值与假手术组无差异;凹入试验中hPTH1-34组Can.Load和 Can.Stiff明显高于去卵巢对照组,和假手术组无差异;三点弯曲实验中hPTH1-34组E.Modulus值与对照组无统计学差异;hPTH1-34 组血ALP和UDpd/Cr水平低于去卵巢对照组,但高于假手术组。结论 hPTH1-34可以明显增加去卵巢骨质疏松大鼠的骨量和 骨生物力学特性。
Objective To investigate the therapeutic effect of human parathyroid hormone 1-34 (hPTH1-34) on osteoporosis in ovariectomized rats. Methods Wister rat hPTH1-34 (40 μg · kg-1 · d-1, sc, 5 times a week) or ovariectomized for 6 weeks or saline for 8 weeks, sham-operated rats were not treated. The lumbar spine 1-6 (L1-6), right femur and blood and urine samples were collected for bone mineral density measurement, bone biomechanics and biochemical determination. Results The BMD of L1-6, distal femur and femur in hPTH1-34 group was significantly higher than that in ovariectomized group, and there was no difference in BMD of L1-6 and femoral shaft between sham operation group and hPTH1-34 group. 34 groups Can. Load and Can. Stiff was significantly higher than the ovariectomized control group, and sham operation group no difference; three point bending test hPTH1-34 group E. There was no significant difference in the values of Modulus between the two groups. The levels of ALP and UDpd / Cr in hPTH1-34 group were lower than those in ovariectomized group but higher than those in sham operation group. Conclusion hPTH1-34 can significantly increase bone mass and biomechanical properties in ovariectomized rats.
