目的:在建立人外周血白细胞蛋白质组分析方法的基础上,对健康志愿者和急性淋巴细胞白血病(ALL)患者白细胞的蛋白质组进行差异性分析,寻找与ALL相关的蛋白质。方法:收集10例健康志愿者和10例ALL患者外周血标本,分离白细胞,提取白细胞蛋白质,双向电泳分离蛋白质,PDQuest2D软件对双向电泳图谱进行差异分析,基质辅助的激光解析飞行时间质谱鉴定差异蛋白质。结果:利用PDQuest2D软件对健康志愿者与ALL患者外周血白细胞蛋白质双向电泳图谱进行差异分析,根据基质辅助的激光解析飞行时间质谱鉴定结果确定了与ALL相关的蛋白质有AnnexinA5、cytoskeleton14等。结论:经质谱鉴定和生物信息学分析,确定了蛋白质AnnexinA5、cytoskeleton14与ALL的发生有着密切的关系,为寻找该类型白血病的药物治疗靶点及早期诊断的分子标志物提供了良好的实验依据。
OBJECTIVE: To establish a method for proteomic analysis of human peripheral leukocytes, and to analyze the differential leukocyte proteome in healthy volunteers and acute lymphoblastic leukemia (ALL) patients in order to find ALL related proteins. Methods: Peripheral blood samples were collected from 10 healthy volunteers and 10 patients with ALL. Leukocytes were isolated and leukocyte proteins were extracted. Proteins were separated by two-dimensional electrophoresis. PDQuest2D software was used to analyze the two-dimensional electrophoresis profiles. Matrix-assisted laser desorption / . Results: PDQuest2D software was used to analyze the difference of two-dimensional electrophoresis patterns of peripheral blood leukocytes between healthy volunteers and ALL patients. AnnexinA5, cytoskeleton14 and other proteins related to ALL were identified according to the results of matrix-assisted laser desorption / ionization time of flight mass spectrometry. Conclusion: The results of mass spectrometry and bioinformatics analysis confirmed that Annexin A5 and cytoskeleton14 are closely related to the occurrence of ALL, and provide a good experimental basis for finding the drug target and early diagnosis molecular marker of leukemia.
