目的:建立超快速液相色谱一光电二极管阵列检测器法(UFLC-DAD)同时测定三叶青药材中原花青素B1和儿茶素的含量。方法:采用菲罗门Kinetex C_(18)色谱柱(4.6 mm×100 mm,2.6μm),流动相为乙腈(B)-0.1%甲酸(A)梯度洗脱,流速0.4 mL·min~1,二极管阵列检测器的检测波长为278 nm,柱温30℃。结果:原花青素B1和儿茶素的线性范围分别为1.25~12.35μg·mL~1(r=0.9992)、1.85~18.28μg·mL~1(r=0.9990),平均加样回收率分别为97.71%(RSD=2.80%)、99.64%(RSD=2.90%)。结论:建立的UFLC-DAD定量分析方法简便、快捷、准确,可用于三叶青药材的质量评价,为三叶青的质量提供实验依据。
OBJECTIVE: To establish an ultra-fast liquid chromatography-photodiode array detector (UFLC-DAD) method for the simultaneous determination of procyanidin B1 and catechin in clover leaves. METHODS: The gradient elution was performed on a Phenomenex Kinetex C_ (18) column (4.6 mm × 100 mm, 2.6 μm) with a mobile phase of acetonitrile-B 0.1% formic acid (A) at a flow rate of 0.4 mL · min -1, Diode array detector detection wavelength of 278 nm, column temperature 30 ℃. RESULTS: The linear range of procyanidin B1 and catechin were 1.25 ~ 12.35μg · mL ~ 1 (r = 0.9992) and 1.85 ~ 18.28μg · mL ~ 1 (r = 0.9990) .The average recoveries were 97.71% (RSD = 2.80%), 99.64% (RSD = 2.90%). Conclusion: The established UFLC-DAD quantitative analysis method is simple, fast and accurate, which can be used to evaluate the quality of clover green herbs and provide the experimental basis for the quality of clover leaf.
