目的 探讨腺相关病毒(adeno-associated virus,AAV)作为载体,介导BDNF基因,转染耳蜗细胞后对顺铂致豚鼠耳蜗损害的保护作用.方法 构建rAAV-BDNF,将人工外淋巴液、rAAV-BDNF微注射到顺铂耳聋模型鼠耳蜗内,经脑干诱发电位检测听力阈值及I波潜伏期,ELISA法检测耳蜗外淋巴液BDNF蛋白表达,耳蜗基底膜铺片检测外毛细胞的凋亡.结果实验组和对照组ABR听阈及I波潜伏期、外毛细胞缺失率、BDNF的表达比较差异有统计学意义.结论 AAV能介导BDNF基因转染耳蜗分泌BDNF,拮抗顺铂豚鼠耳蜗毒性,为感音神经性聋的基因治疗提供依据.“,”Objective To observe the otoprotective efficacy of adeno-associated virus (AAV) as vector mediated brain derived neurotrophic factor(BDNF) gene on cisplatin-induced ototoxicity for cochlear gene therapy. Methods rAAV-BDNF was constructed,and artificial perilymph,rAAV-BDNF were microinjected into the cochlea respectively.The cochlear function was analyzed with evoked auditory brainstem responses(ABR).The BDNF expression was detected with ELISA,and outer hair cells(OHC) were checked for evaluation.Results There was a statistical significance of difference between the two groups in auditory ability, OHC loss,and expression of BDNF.Conclusion AAV-mediated BDNF expression can preserve the cochlear function from the cisplatin-induced ototoxicity.
