目的研究亚低温对易卒中型肾血管性高血压大鼠(RHRSP)复制的大脑中动脉闭塞(MCAO)模型的梗死灶大小及对BCL-2蛋白表达的影响。方法用环型银夹以双肾双夹法复制RHRSP,改良Longa’s线栓法制成MCAO缺血模型,冰袋法诱导亚低温,TTC染色测定梗死灶体积,免疫组化法、图象识别系统计算BCL-2蛋白表达强度。结果1亚低温能明显缩小梗死灶体积;2亚低温组BCL-2蛋白表达明显强于正常体温组。结论亚低温增强缺血后基因BCL-2的表达可能是它具有缺血脑保护作用机制之一。
Objective To investigate the effects of mild hypothermia on the infarct size and the expression of BCL-2 protein in middle cerebral artery occlusion (MCAO) model of rats with stroke-induced renovascular hypertension (RHRSP). Methods RHRSP was duplicated with ring-shaped silver clip with twin-kidney method, MCAO model with modified Longa’s thread method, and infarct size with immunohistochemistry and image recognition system BCL-2 protein expression intensity was calculated. Results 1 mild hypothermia can significantly reduce infarct volume; 2 mild hypothermia group BCL-2 protein expression was significantly stronger than the normal body temperature group. CONCLUSION: Mild hypothermia enhances the expression of BCL-2 gene after ischemia may be one of the mechanisms of its protective effects on cerebral ischemia.
