目的 :研究氧化砷 (As2 O3)对白血病细胞系 NB4 、Jurkat细胞周期的影响及对细胞周期及凋亡相关蛋白的调节作用。方法 :以 MTT、基因组 DNA电泳、蛋白 / DNA双参数流式细胞术等方法研究 As2 O3对白血病细胞的作用及机制。结果 :随着 As2 O3作用时间的延长 ,白血病细胞增殖明显受到抑制 ,DNA电泳出现“梯”状条带 ,流式细胞术检出亚 G1 峰 ,细胞周期阻滞于 G1 和 G2 / M期 ,部分细胞周期及凋亡相关的调控蛋白表达改变。结论 :As2 O3通过改变细胞周期及凋亡相关蛋白的表达而抑制白血病细胞增殖和诱导其凋亡
AIM: To investigate the effects of arsenic trioxide (As2O3) on the cell cycle of leukemic cell lines NB4 and Jurkat and the regulation of cell cycle and apoptosis related proteins. Methods: MTT, genomic DNA electrophoresis, protein / DNA dual-parameter flow cytometry and other methods As2 O3 on leukemia cells and its mechanism. RESULTS: The proliferation of leukemia cells was significantly inhibited with the prolongation of As 2 O 3 treatment. The “ladder” bands were observed by DNA electrophoresis. The sub G1 peak was detected by flow cytometry. The cell cycle was arrested at G1 and G2 / M phases. Part of the cell cycle and apoptosis-related regulatory protein expression changes. Conclusion: As2 O3 can inhibit the proliferation and induce the apoptosis of leukemia cells by altering the expression of cell cycle and apoptosis-related proteins
