应用荧光原位杂交和基因组原位杂交技术进行rDNA在椭圆悬钩子、红毛悬钩子和栽秧泡中期染色体上的定位及3个物种亲缘关系研究。结果表明:3个物种均具有1对45S rDNA位点,位于染色体短臂端部;椭圆悬钩子和红毛悬钩子的1对5S rDNA位点位于染色体短臂近着丝粒处,栽秧泡的1对5S rDNA位于染色体短臂端部。基因组原位杂交表明,椭圆悬钩子与红毛悬钩子之间共有的重复序列主要分布于14条染色体近着丝粒区或端部,杂交信号较强;而与栽秧泡之间共享的重复序列仅存在8条染色体近着丝粒区或端部,且杂交信号相对较弱。红毛悬钩子与处于绒毛叶亚组的茅莓关系较近,与处于柔毛叶亚组的插田泡关系较远。结合形态特征的差异,支持将椭圆悬钩子和红毛悬钩子归并,并将归并后的椭圆悬钩子放在绒毛叶亚组;将栽秧泡从椭圆悬钩子的变种独立、提升为种,仍留在绒毛叶亚组。
Fluorescence in situ hybridization and genomic in situ hybridization (rDNA) were used to determine the rDNA loci on the chromosomes of midges of Erythroculter, Erigeron brevis and Transplanting seedlings and the genetic relationship among the three species. The results showed that all the three species had a pair of 45S rDNA loci located at the ends of the short arm of the chromosome. One pair of 5S rDNA loci located on the short arm of the chromosome, A pair of 5S rDNA is located at the short arm end of the chromosome. Genomic in situ hybridization showed that the repeat sequences shared between Rubus and Rubus were mainly distributed in the proximal centromere region or the end of 14 chromosomes and the hybridization signal was strong. There are only eight chromosome centromeric regions or ends in the sequence, and the hybridization signal is relatively weak. The red-haired raspberry has a close relationship with the red-haired raspberry in the sub-group of velvetleaf, and has a long relationship with the frog in the sub-group of the soft-haired sub-group. Combined with the morphological differences, support the oval and red hair hook rubus merge, and the merged elliptical rubble on the villus leaf subgroup; the planting seedlings from oval Rubus variants independent, promoted to species, still Stay in the villus leaves subgroup.
