从人低分化鼻咽癌细胞系(CNE-2Z)中分离出单克隆株4-2-H5、5-1-F_1、5-2-F_7进行体外培养,分别计算各克隆株细胞倍增时间和分裂指数,用玻璃微电极细胞内记录方法测量细胞膜电位。结果表明:(1)随培养时间延长,三个克隆株细胞膜电位负值都减小,差异有显著意义;(2)膜电位负值下降率与细胞株生长增殖能力呈正相关,(3)H_5,F_1细胞膜电位负值与细胞外K ̄+浓度的对数值接近反比关系。提高培养液Na ̄+浓度。h_5、f_1细胞膜电位负值稍下降,前者无显著性意义,后者有显著性意义,提示H_5、F_1细胞膜电位主要与K ̄+外流有关,在F_1细胞,膜电位形成还与Na ̄+有一定关系。
The monoclonal strains 4-2-H5, 5-1-F_1, 5-2-F_7 were isolated from human poorly differentiated nasopharyngeal carcinoma cell line (CNE-2Z) and cultured in vitro. The cell doubling time and Splitting index, cell membrane potential was measured using a glass microelectrode intracellular recording method. The results showed that: (1) the negative value of cell membrane potential of all three clones decreased with the prolongation of culture time, the difference was significant; (2) the negative rate of membrane potential was positively correlated with cell proliferation and proliferation; (3) H_5 , F_1 cell membrane potential negative and extracellular K ~ + concentration logarithmically inversely proportional relationship. Increase the concentration of Na ~ + in culture solution. h_5, f_1 cell membrane potential decreased slightly, the former was not significant, the latter has a significant meaning, suggesting that H_5, F_1 cell membrane potential and K ~ + efflux, in F_1 cells, the membrane potential is also associated with Na ~ + A certain relationship.
