目的:研究化浊行血颗粒对动脉粥样硬化(AS)模型大鼠血脂和颈总动脉结构的影响,探讨治疗AS的有效中药。方法:实验动物按体质量随机分为正常对照组、模型组和药物组,每组12只,采用高脂饲料法建立大鼠食饵性AS模型。建模后,药物组灌胃给予化浊行血颗粒水溶液(12g生药·kg-1·d-1),正常对照组、模型组分别给予等容量蒸馏水,连续8周。观察血脂和颈总动脉结构变化。结果:血脂:与正常对照组比较,模型组血清胆固醇(CH)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)明显增高(P<0.01)。药物组与模型组比较,血清CH、TG、LDL-C明显降低(P<0.01),高密度脂蛋白胆固醇(HDL-C)明显增高(P<0.05)。颈总动脉结构:正常对照组颈总动脉结构正常;模型组内膜明显增厚,有大量脂滴沉积和胆固醇结晶形成;药物组内膜较模型组变薄,脂滴沉积和胆固醇结晶减少。结论:化浊行血颗粒可有效控制模型大鼠血脂水平,能稳定AS斑块、保护血管内膜、减少脂滴沉积和胆固醇结晶形成,对抗AS内膜增厚和斑块形成。
Objective: To study the effect of Huazhuo xuexue granule on the structure of blood lipid and common carotid arteries in rats with atherosclerosis (AS) and explore the effective traditional Chinese medicine for treating AS. Methods: The experimental animals were randomly divided into normal control group, model group and drug group according to the body weight. Twelve rats in each group were fed with high-fat diet to establish the rat model of pretension of AS. After modeling, the drug group was given intragastric administration of Huazhuo blood granules aqueous solution (12g crude drug · kg-1 · d-1), normal control group, model group were given the same volume of distilled water for 8 weeks. The changes of blood lipids and common carotid arteries were observed. Results: Compared with normal control group, serum cholesterol, triglyceride and low density lipoprotein cholesterol (LDL-C) in model group were significantly increased (P <0.01). Compared with model group, serum CH, TG and LDL-C in the drug group were significantly lower (P <0.01) and HDL-C significantly higher (P <0.05). The structure of common carotid artery: The structure of common carotid artery in normal control group was normal. The intima of model group was obviously thickened with a large number of lipid droplets deposition and cholesterol crystal formation. The inner membrane of drug-treated group was thinner than that of model group, and lipid droplets deposition and cholesterol crystallization decreased. Conclusion: Huazhuo xuexue granules can effectively control the level of blood lipid in model rats, stabilize AS plaque, protect the intima of blood vessels, decrease the deposition of lipid droplets and the formation of cholesterol crystals, and resist AS intimal thickening and plaque formation.
