目的:探讨金丹活性部位对心肌缺血再灌注损伤(MIRI)大鼠心肌Bax、Bcl-2蛋白表达的影响。方法:将金丹活性部位对MIRI大鼠进行干预,应用免疫组化法检测大鼠心肌Bax、Bcl-2表达的变化。结果:与模型组相比,水洗脱组,60%醇洗脱组和预处理组Bax灰度值均有明显升高;Bcl-2灰度值均明显下降(P<0.05,P<0.01)。结论:金丹60%乙醇洗脱组与水洗脱组均能明显上调Bcl-2蛋白表达,下调Bax的蛋白表达,说明其可能通过抑制细胞凋亡而发挥保护MIRI的作用。
Objective: To investigate the effect of saponins active site on myocardial Bax and Bcl-2 protein expression in myocardial ischemia-reperfusion injury (MIRI) rats. Methods: The active site of saponin was used to intervene in MIRI rats. The expression of Bax and Bcl-2 in myocardium of rats were detected by immunohistochemistry. Results: Compared with the model group, the gray value of Bax in water elution group, 60% alcohol elution group and pretreatment group were significantly increased; the gray value of Bcl-2 was significantly decreased (P <0.05, P <0.01 ). CONCLUSION: The expression of Bcl-2 protein and the protein expression of Bax were significantly up-regulated by 60% ethanol elution group and water elution group, indicating that it may play a role in protecting MIRI by inhibiting apoptosis.
