米非司酮对小鼠卵丘颗粒细胞超微结构的影响

目的:观察米非司酮对小鼠卵丘颗粒细胞超微结构的影响。方法:雌性昆明小鼠40只,随机均分为4组:对照组(A组),米非司酮0.048mg/kg组(B组)、0.143mg/kg组(C组)、0.429mg/kg组(D组)。4组动物均于动情前期一次灌胃给药,10h后取其双侧卵巢组织,透射电镜观察4组小鼠成熟卵泡内卵丘颗粒细胞超微结构的变化。结果:与A组相比,B组小鼠卵丘颗粒细胞的间隙变宽,部分线粒体水肿明显。C组细胞外形不规则,细胞间隙宽,可见髓样小体,线粒体更加肿胀,呈空泡状。D组细胞胞浆内有凋亡现象出现,胞浆内可见部分凋亡小体,细胞核形态不规则。结论:米非司酮可损伤小鼠成熟卵泡中卵丘颗粒细胞超微结构,随着剂量的增加,凋亡现象明显。 Objective: To observe the effect of mifepristone on the ultrastructure of cumulus granulosa cells in mice. Methods: Forty female Kunming mice were randomly divided into four groups: control group (A group), mifepristone 0.048mg / kg group (B group), 0.143mg / kg group (C group), 0.429mg / kg group (group D). Four groups of animals were intragastrically administered once a day before estrus. After 10 hours, the ovaries were taken and the ultrastructure of cumulus granulosa cells in the mature follicles of the four groups were observed under transmission electron microscope. Results: Compared with group A, the gap of cumulus granulosa cells in group B was broadened and some mitochondria edema was obvious. C group of irregular cell shape, cell gap wide, visible medullary bodies, mitochondria more swelling, vacuolization. In group D, apoptosis appeared in the cytoplasm, some apoptotic bodies were seen in the cytoplasm, and the nuclear morphology was irregular. Conclusion: Mifepristone can damage the ultrastructure of cumulus granulosa cells in mature follicles in mice, with the dose increasing, the apoptosis phenomenon is obvious.
