目的 探讨TNFRp55和TNFRp75在各种肿瘤组织中的表达与新型rhTNF对肿瘤细胞生长的抑制作用的关系。方法应用免疫组织化学染色法,观察120例肿瘤组织和10株体外培养的肿瘤细胞中,TNFRp55和TNFRp75的表达及分布。同时应用结晶紫摄入法,观察nrhTNF对肿瘤细胞生长的抑制。结果 在肿瘤组织和肿瘤细胞中,TNFRp55的阳性表达率分别为82.5%和99%,TMFRp75分别为 84.2%和98.8%,不同类型肿瘤间的表达无明显差异(P>0.05).nrhTNF可抑制SW480、SGC7901、8910SMMC7721和BEL7402这5株肿瘤细胞生长,但是不能抑制另外5株肿瘤细胞A549、PLA801、Hela、Eca109和GRC-l的生长。结论TNFR广泛存在于各种肿瘤细胞上,nrhTNF抑制肿瘤细胞生长的程度与TNFRp55的表达率无相关性。
Objective To investigate the relationship between the expression of TNFRp55 and TNFRp75 in various tumor tissues and the inhibitory effect of rhTNF on tumor cell growth. Methods Immunohistochemical staining was used to observe the expression and distribution of TNFRp55 and TNFRp75 in 120 tumor tissues and 10 in vitro cultured tumor cells. At the same time, crystal violet intake method was used to observe the inhibition of tumor growth by nrhTNF. Results The positive expression rates of TNFRp55 in tumor tissues and tumor cells were 82.5% and 99%, respectively, and those of TMFRp75 were 84.2% and 98.8% respectively. There was no significant difference between different types of tumors (P> 0 .05). nrhTNF inhibited the growth of 5 tumor cells, SW480, SGC7901, 8910SMMC7721 and BEL7402, but did not inhibit the growth of the other 5 tumor cells, A549, PLA801, Hela, Eca109 and GRC-1. Conclusion TNFR exists in a wide range of tumor cells. The degree of tumor growth inhibition by nrhTNF is not correlated with the expression of TNFRp55.
