目的:探讨熊去氧胆酸对肝硬化大鼠血清IL-6、MMP-3、TIMP-1水平及胃肠Cajal细胞c-kit表达的影响。方法:选取72只大鼠,随机分为四组,每组18只。空白组给予正常饲养,其余三组均注射CCl_4予酒精和高蛋白饮食,熊去氧胆酸组大鼠注射熊去氧胆酸1.5 mg/Kg·d;吗丁啉组给予吗丁啉0.85 mg/Kg·d;模型组仅给予注射生理盐水。治疗结束后,对比各组血清IL-6、MMP-3、TIMP-1水平及胃肠Cajal细胞c-kit含量。结果:与空白组比较,模型组大鼠血清IL-6、MMP-3、TIMP-1水平均明显升高(P<0.01),胃肠Cajal细胞c-kit含量显著降低(P<0.01)。与模型组相比,熊去氧胆酸组与吗丁啉组大鼠血清IL-6、MMP-3、TIMP-1水平均显著降低(P<0.01),小肠推进率增高(P<0.01),胃肠Cajal细胞c-kit含量显著升高(P<0.01);与吗丁啉组相比,熊去氧胆酸组血清IL-6、MMP-3、TIMP-1水平较低(P<0.01),小肠推进率、胃肠Cajal细胞c-kit含量显著升高(P<0.01)。结论:熊去氧胆酸对肝硬化具有较好疗效,可改善肝纤维化和胃肠功能,这可能与其降低血清IL-6、MMP-3、TIMP-1水平及提高胃肠Cajal细胞C-kit含量有关。
Objective: To investigate the effects of ursodeoxycholic acid on serum IL-6, MMP-3, TIMP-1 and c-kit expression in gastrointestinal Cajal cells in cirrhotic rats. Methods: Seventy-two rats were selected and randomly divided into four groups with 18 rats in each group. The blank group was given normal feeding, the other three groups were injected with CCl_4 to alcohol and high protein diet, the ursodeoxycholic acid group was injected with ursodeoxycholic acid 1.5 mg / Kg · d, the domperidone group was given domperidone 0.85 mg / Kg · d; model group was given only saline injection. After the treatment, the levels of IL-6, MMP-3, TIMP-1 in serum and the c-kit content in Cajal cells were compared. Results: Compared with the blank group, the levels of IL-6, MMP-3 and TIMP-1 in the model group were significantly increased (P <0.01) and the content of c-kit in the Cajal cells was significantly decreased (P <0.01). Compared with the model group, the levels of IL-6, MMP-3 and TIMP-1 in the ursodeoxycholic acid group and the domperidone group were significantly decreased (P <0.01), and the intestinal propulsion rate was increased (P <0.01) (P <0.01). Compared with the control group, the levels of IL-6, MMP-3 and TIMP-1 in the ursodeoxycholic acid group were lower (P < 0.01), small intestine propulsion rate and c-kit content in gastrointestinal Cajal cells were significantly increased (P <0.01). Conclusion: Ursodeoxycholic acid has a good effect on liver cirrhosis, can improve liver fibrosis and gastrointestinal function, which may be related to its lower levels of serum IL-6, MMP-3, TIMP-1 and improve gastrointestinal Cajal cells C- kit content.
