采用“搓网”方法将8种96例不同的恶性肿瘤手术切除标本制成细胞膜完整的单细胞悬液,并从6例癌性腹水中提取富含癌细胞的悬液。用流式细胞免疫学方法检测癌细胞(MDR1)表达产物P170含量。经上述方法制成悬液中的癌细胞丰富、荧光显微镜下观察胞膜完整,P170阳性细胞抗体荧光位于胞膜上。本文认为用流式细胞免疫学技术检测恶性实体瘤细胞MDR1表达产物P170含量简便易行,不仅可以了解肿瘤细胞始基的药物敏感性,还可在化疗过程中动态监测药物敏感性的变化。此法有明确的临床价值
Eight kinds of 96 different surgical specimens of malignant tumors were made into single-cell suspensions with intact cell membranes using the “mesh net” method, and a suspension rich in cancer cells was extracted from 6 cases of cancerous ascites. The P170 content of cancer cell (MDR1) expression product was detected by flow cytometric immunoassay. After the above method, the cancer cells prepared in the suspension were abundant, and the cell membrane was observed under the fluorescence microscope. The fluorescence of the P170 positive cell antibody was located on the cell membrane. In this paper, the use of flow cytometric immunoassay to detect the P170 content of MDR1 expression product in malignant solid tumor cells is simple and feasible, not only can understand the drug sensitivity of the tumor cell primordia, but also dynamic monitoring of drug sensitivity changes during chemotherapy. This method has a clear clinical value
