目的 :探讨益气通络方对神经元缺氧凋亡的作用 ,以及对神经元缺氧过程氧自由基 (OFR)、一氧化氮 (NO)生成和bcl 2基因表达的影响。方法 :采用Daniel方法建立神经元缺氧凋亡模型 ,加入益气通络方药物血清 ,应用碘化丙啶 (PropidiumiodidePI)染色法经流式细胞仪检测神经元凋亡率 ,分光光度法检测MDA、NO浓度 ,免疫组化法检测bcl 2表达。结果 :益气通络方显著抑制缺氧导致的神经元凋亡 ,并减少神经元缺氧过程NO、OFR的生成 ,上调bcl 2基因的表达。结论 :益气通络方对神经元缺氧凋亡有抑制作用 ,其机制可能与其减少神经元缺氧过程NO、OFR生成 ,及上调bcl 2基因表达有关。
Objective: To investigate the effect of Yiqi Tongluo Fang on neuronal apoptosis and the effect of oxygen radical (OFR), nitric oxide (NO) production and bcl 2 gene expression during neuronal hypoxia. METHODS: The neuronal apoptosis model was established by Daniel method. Serum of Yiqi Tongluo prescription was added. Propidiumiodide PI staining was used to detect the apoptosis rate of neurons by flow cytometry. Spectrophotometric method was used to detect MDA. , NO concentration, immunohistochemical method to detect bcl 2 expression. Results: YQT significantly inhibited neuronal apoptosis induced by hypoxia, reduced the production of NO and OFR during hypoxia, and up-regulated the expression of bcl 2 gene. Conclusion : Yiqi Tongluo Prescription can inhibit the apoptosis of neurons induced by hypoxia. Its mechanism may be related to the decrease of NO, OFR production and the up-regulation of bcl 2 gene expression in hypoxic neurons.
