目的研究HIV/AIDS患者外周血Th9细胞及其转录因子PU.1的表达量,探讨其与艾滋病疾病进展的相关性。方法40例HIV/AIDS患者根据CD4~+T淋巴细胞分为A、B 2组(A组CD4~+T淋巴细胞<350,B组CD4~+T淋巴细胞>350),同时,选取20例健康人为对照组,采用流式细胞技术及RT-PCR技术检测其外周血CD4~+T淋巴细胞亚群(Th1、Th2、Th9)及其相关转录因子(T-bet、GATA3、PU.1)的表达量,通过比较3组间Th9细胞及其转录因子PU.1表达量的差异,分析Th9细胞及其转录因子与艾滋病疾病进展的相关性。结果与健康人群相比,HIV/AIDS患者CD4~+T细胞绝对计数与百分比明显降低,CD8~+T细胞的绝对计数与百分比明显升高;3组组间比较显示,Th1细胞百分比A组(6.71±3.05)和B组(5.82±2.52)低于健康对照组(10.89±6.82)差异有统计学意义(P<0.05);Th2细胞百分比3组间差异无统计学意义;Th9细胞百分比A组(2.0±0.69)、B组(0.66±0.23)和健康对照组(0.40±0.16)之间差异有统计学意义(P<0.05);转录因子PU.1相对表达量A组(3.30±0.78)和B组(1.59±0.44)之间差异有统计学意义(P<0.05)。Th9细胞及其转录因子PU.1相对表达量与CD4~+T细胞成负相关(r=-0.637,r=-0.746,P=0.000)。结论 HIV/AIDS患者外周血Th9细胞及其转录因子PU.1的表达量随着疾病进展逐渐升高,两者与艾滋病疾病进展密切相关,可作为判断疾病进展的指标。

The comparison of the three groups showed that the percentages of Th1 cells in group A 6.71 ± 3.05) and group B (5.82 ± 2.52), which were lower than that of healthy control group (10.89 ± 6.82) (P <0.05). There was no significant difference in the percentage of Th2 cells between the three groups. The percentage of Th9 cells in group A (2.0 ± 0.69), group B (0.66 ± 0.23) and healthy control group (0.40 ± 0.16) (P <0.05). The relative expression of transcription factor PU.1 in group A was (3.30 ± 0.78) And group B (1.59 ± 0.44), the difference was statistically significant (P <0.05). The relative expression level of Thl cells and its transcription factor PU.1 was negatively correlated with CD4 ~ + T cells (r = -0.637, r = -0.746, P = 0.000). Conclusion The expression of Th9 cells and the transcription factor PU.1 in peripheral blood of HIV / AIDS patients gradually increases with the progression of the disease. Both of them are closely related to the progression of AIDS and can be used as an index to judge the progress of the disease.
