目的用分子生物学方法研究小盾纤恙螨的媒介意义。方法聚合酶链反应和原位分子杂交法。结果从恙螨体内检测到肾综合征出血热病毒RNA;取恙螨幼虫、若虫定期做肾综合征出血热病毒TCID50/ml滴定,证明恙螨体内肾综合征出血热病毒可传播并有增殖现象;用原位分子杂交技术在恙螨的卵巢细胞、支肠细胞等组织中检测到肾综合征出血热病毒RNA。结论研究结果为恙螨的媒介意义提供了具有分子水平的直接证据,对肾综合征出血热的流行病学和预防具有重要的理论意义和实用价值。
Objective To study the mediaelement of chigger mite by molecular biology method. Methods Polymerase chain reaction and in situ hybridization. Results Detection of hemorrhagic fever with renal syndrome virus RNA from chigger mites; taking chigger larvae and nymphs regularly for TCID50 / ml titration of hemorrhagic fever with renal syndrome, demonstrating that chigger mite hemorrhagic fever with renal syndrome hemorrhagic fever virus can spread and proliferate In situ hybridization was used to detect hemorrhagic fever with renal syndrome virus RNA in chigger mite ovary cells and colonic cells. Conclusion The findings provide direct evidence of the molecular significance of chigger mite and have important theoretical and practical value for the epidemiology and prevention of hemorrhagic fever with renal syndrome.
