目的:探讨心脉康胶囊对大鼠缺血再灌注所致心肌梗死的作用机制。方法:将48只雄性Wistar大鼠随机分为假手术组、模型组、合心爽组和心脉康高、中、低剂量组,每组8只。对除假手术组的另外5组大鼠进行缺血再灌注所致心肌梗死模型制备。通过十二指肠给药,合心爽组剂量为16mg/kg体质量,心脉康高、中、低剂量组给药剂量分别为4.0、2.0、1.0g/kg体质量,给药体积为3mL/kg,假手术组和模型组不予给药。各组大鼠恢复自主呼吸2h后,经腹主动脉取血,用比色法测定其血清超氧化物歧化酶(SOD)和丙二醛(MDA)含量;采用多媒体彩色病理图文分析系统,以固定象距测量正常心肌与梗死心肌面积,观察心肌梗死程度。结果:心脉康胶囊各剂量组能明显减轻大鼠缺血再灌注所致心肌梗死程度,缩小梗死面积,减轻梗死区重量,同时增加SOD活性,与模型组比较,差异均有统计学意义(P<0.05,P<0.01);各药物组MDA含量与模型组比较无显著变化。结论:心脉康胶囊对大鼠心肌缺血再灌注损伤的保护作用可能是通过抑制脂质过氧化、减少氧自由基对心肌的损伤来实现的。
Objective: To investigate the mechanism of Xinmaikang capsule on myocardial infarction induced by ischemia-reperfusion in rats. Methods: Forty eight male Wistar rats were randomly divided into sham operation group, model group, Hexin group and Xinmaikang high, middle and low dose group, with 8 rats in each group. Myocardial infarction models were prepared by ischemia-reperfusion in another 5 groups of rats except the sham operation group. Through the duodenum, the volume of Qixinshu group was 16mg / kg body weight, the dosage of Xinmaikang high, middle and low dose groups were 4.0, 2.0, 1.0g / kg body weight respectively, 3mL / kg, sham operation group and model group were not given. The rats in each group recovered spontaneously breathing for 2h, then the blood was taken from the abdominal aorta, and the levels of serum superoxide dismutase (SOD) and malondialdehyde (MDA) were measured by colorimetric method. By using the multimedia color pathological image analysis system, The normal myocardial infarction area and myocardial infarction area were measured by fixed image distance, and the degree of myocardial infarction was observed. Results: Each dose of Xinmaikang capsule could relieve the degree of myocardial infarction induced by ischemia-reperfusion, reduce the infarct size, reduce the weight of infarction area and increase the activity of SOD. Compared with the model group, the differences were statistically significant ( P <0.05, P <0.01). There was no significant difference in MDA content in each drug group compared with model group. Conclusion: The protective effect of Xinmaikang capsule on myocardial ischemia-reperfusion injury may be through inhibiting lipid peroxidation and reducing the damage of oxygen free radicals to myocardium.
