目的研究解郁安神方对慢性温和应激(CMS)抑郁动物模型学习记忆功能的影响。方法按照体重将成年雄性昆明小鼠随机分为5组(每组6只):空白组,模型组,大中小3个剂量实验组(解郁安神方,50,100,200 mg·kg~(-1))。制备CMS抑郁大鼠模型。用Morris水迷宫(MWM)评价实验动物空间学习记忆功能。结果给药后第4天,模型组与小中大3个剂量实验组平台潜伏期分别是(42.33±7.76)s与(27.83±6.34),(17.00±6.42),(12.67±5.28)s,与模型组比较,3个剂量实验组差异均有统计学意义(均P<0.05),提示解郁安神方醇提物可改善CMS抑郁动物模型学习功能。给药后第5天,模型组与大剂量实验组平台象限潜伏期分别是(48.67±13.37),(24.33±15.33)s,与模型组比较,大剂量实验组差异有统计学意义(P<0.05)。模型组与中大2个剂量实验组平台象限时间分别是(38.17±11.36)s与(64.17±8.84),(65.00±9.75)s,与模型组比较,2个剂量实验组差异均有统计学意义(均P<0.05),提示解郁安神方醇提物可改善CMS抑郁动物模型记忆功能。结论解郁安神方可以影响CMS抑郁动物模型MWM定位航行训练与空间探索测试不同阶段行为学变化,提示其可改善CMS抑郁动物模型学习记忆功能。
Objective To study the effects of Jieyu Anshen recipe on the learning and memory abilities in chronic mild stress (CMS) depression animal model. Methods Male Kunming mice were randomly divided into 5 groups (6 rats in each group) according to body weight: blank group, model group, medium, small and medium dose groups (Jieyu Anshen Fang, 50,100,200 mg · kg -1) . Preparation of CMS Depression Rat Model. Morris water maze (MWM) was used to evaluate the spatial learning and memory function of experimental animals. Results On the 4th day after the administration, the latency of platform in the three experimental groups was (42.33 ± 7.76) s and (27.83 ± 6.34), (17.00 ± 6.42) and (12.67 ± 5.28) s, respectively Compared with the model group, the differences of the three dose groups were statistically significant (all P <0.05), suggesting that Jieyu Anshen Fang alcoholic extract can improve the learning function of CMS depression model. On the fifth day after administration, the latency of platform quadrant in model group and high-dose experimental group was (48.67 ± 13.37) and (24.33 ± 15.33) s, respectively. Compared with model group, the experimental group had significant difference (P <0.05 ). The quadrant time of the experimental group and the experimental group were (38.17 ± 11.36) s and (64.17 ± 8.84) and (65.00 ± 9.75) s, respectively. Compared with the model group, the differences of the two experimental groups were statistically significant (All P <0.05), suggesting that Jieyu Anshen Fang alcohol extract can improve the memory function of CMS depression model. Conclusions JieYuShenJingFang can affect the behavioral changes in different stages of MWM positioning navigation training and space exploration test in CMS depressive animal model, suggesting that it can improve the learning and memory function of CMS depression model.
