制作由大鼠单侧输尿管梗阻所致的肾间质纤维化动物模型。以α-平滑肌肌动蛋白(α-SMA)作为肌纤维母细胞(MFB)的特异性标志物,观察肾小管上皮细胞及间质内MFB的表达情况。15只SD大鼠随机分为注射和口服红景天组及实验对照组。用药治疗10天后鼠全部处死。观察病肾HE、PASM及免疫组化α-SMA染色。结果:在实验结束时对照组肾小管上皮细胞及间质α-SMA表达明显增加。而应用红景天组α-SMA的表达却明显减少(P<0.05),口服红景天组则效果更好。表明:α-SMA是MFB较好的标志物。MFB在UUO导致肾间质纤维化中扮演了重要角色。红景天具有抗肾间质纤维化的作用。
An animal model of renal interstitial fibrosis caused by unilateral ureteral obstruction in rats was prepared. Using α-smooth muscle actin (α-SMA) as a specific marker of myofibroblast (MFB), the expression of MFB in renal tubular epithelial cells and interstitium was observed. Fifteen SD rats were randomly divided into injection and oral Rhodiola group and experimental control group. After 10 days of medication, all rats were sacrificed. HE, PASM and immunohistochemical α-SMA staining were observed in diseased kidneys. RESULTS: At the end of the experiment, the expression of α-SMA in renal tubular epithelial cells and interstitium increased significantly in the control group. However, the expression of α-SMA in the Rhodiola group was significantly decreased (P<0.05), and the Rhodiola group was more effective. It shows that α-SMA is a better marker of MFB. MFB plays an important role in UUO leading to renal interstitial fibrosis. Rhodiola has anti-renal interstitial fibrosis.
