筛选出适宜小麦转化的优良基因型扬麦 15 8和扬麦 10号。以扬麦 15 8幼胚愈伤组织为受体 ,利用基因枪介导法将含有小麦黄花叶病毒复制酶基因 (WYMV- N ib8)的 pubi Nib8质粒转入了小麦。T0 代经 PCR扩增分析 ,获得了 14株含 WYMV- N ib8基因的转化植株 ,转化率为 0 .4 3%。T1、T2 代跟踪分子检测和 T2 代田间病毒抗性鉴定结果表明 ,获得了 4个抗 WYMV的转基因株系
The excellent genotypes Yangmai 15 8 and Yangmai 10 were screened for suitable wheat transformation. The immature embryo callus from Yangmai 15 8 was used as a receptor, and the plasmid pMiNib8 containing the wheat yellow mosaic virus replicase gene (WYMV-N ib8) was transferred into wheat using particle bombardment. T0 generation by PCR amplification analysis, obtained 14 WYMV-N ib8 transgenic plants, the conversion rate of 0.43%. The results of T1 and T2 generation of tracking molecular detection and T2 generation of field virus resistance showed that four transgenic lines resistant to WYMV
