目的探讨人参皂苷Compound K(CK)对慢性粒细胞白血病K562细胞周期的影响及机制。方法应用MTT法检测CK对K562细胞增殖的影响,用流式细胞仪分析细胞周期,用RT-PCR分析CyclinB1、cdc2、Survivin基因mRNA变化,用Western-blot法分析CyclinB1、p34cdc2、Survivin蛋白变化。结果人参皂苷CK能显著抑制K562细胞的增殖,呈浓度依赖性,细胞阻滞于G2/M期,cdc2基因mRNA表达下调,CyclinB1、p34cdc2、Survivin蛋白质表达均下调。结论人参皂苷CK能使K562细胞阻滞于G2/M期,该效应可能与CK抑制CyclinB1、cdc2基因表达有关,Survivin蛋白表达下调可能是CK抑制K562细胞周期的关键性分子。
Objective To investigate the effect and mechanism of ginsenoside K (CK) on the cell cycle of chronic myeloid leukemia K562 cells. Methods MTT assay was used to detect the effect of CK on the proliferation of K562 cells. The cell cycle was analyzed by flow cytometry. The mRNA expressions of CyclinB1, cdc2 and Survivin were analyzed by RT-PCR. The protein expressions of CyclinB1, p34cdc2 and Survivin were analyzed by Western-blot. Results Ginsenoside CK could significantly inhibit the proliferation of K562 cells in a concentration-dependent manner. The cell cycle arrest was at G2 / M phase. The expression of cdc2 mRNA was down-regulated while the protein expression of CyclinB1, p34cdc2 and Survivin were down-regulated. Conclusions Ginsenoside CK can arrest K562 cells in G2 / M phase, which may be related to CK inhibiting the expression of CyclinB1 and cdc2 genes. The down-regulation of Survivin protein may be the key factor of CK inhibiting K562 cell cycle.
