目的:研究芪玉三龙汤对荷肺癌小鼠的抑瘤效应及对小鼠肺脏的影响。方法:采用Lewis肺癌细胞株培养移植法建立肺癌荷瘤小鼠模型,随机分为模型组、化疗组、芪玉三龙汤高、中、低剂量组、联合组,每组8只;实验第11天开始芪玉三龙汤低、中、高剂量组小鼠分别按(20.12、40.24、80.48)g/(kg·d)灌胃给药,化疗组按照0.4 m L顺铂腹腔注射给药,联合组以高剂量中药灌胃和顺铂腹腔注射联合给药,模型组按照等量生理盐水给药,1次/d,连续给药10 d;称取瘤质量,计算抑瘤率;实时荧光定量PCR法(RT-q PCR)检测肿瘤组织PI3K mRNA的转录水平;蛋白质免疫印迹法(Western Blot)检测肿瘤组织PI3K p85α、PI3K p110α蛋白水平;免疫组化检测肿瘤组织VEGF蛋白表达,HE染色法观察小鼠肺脏的病理形态变化。结果:与模型组比较,中药高剂量组能够抑制肺癌移植瘤生长,抑瘤率为23.86%,联合组与化疗组抑瘤率相当;芪玉三龙汤能够明显降低肿瘤组织PI3K mRNA的转录水平和PI3K p85α、PI3K p110α蛋白水平,与顺铂联用有一定协同增效作用;并且能够降低VEGF阳性细胞率;光镜下观察肺脏示芪玉三龙汤高、中剂量组以及联合组小鼠肺组织内未见肿瘤转移灶。结论:芪玉三龙汤可温和抑制肺癌移植瘤生长,可能与降低PI3K mRNA转录水平、PI3K p85α、PI3K p110α蛋白表达有关,并可能通过降低VEGF表达减少肺脏的肿瘤转移。

CONCLUSIONS: SDJT can moderately inhibit the growth of lung cancer xenografts and may be related to the decrease of PI3K mRNA expression, PI3K p85α and PI3K p110α protein expression, and may decrease the metastasis of lung cancer by decreasing VEGF expression.
