目的:建立人血浆中多西紫杉醇的高效液相色谱(HPLC)测定方法。方法:取血浆样品500μL,以地西泮为内标,用1mol·L-1碳酸氢钠碱化后乙醚提取,取上清水浴挥干,150μL流动相溶解,取20μL进行HPLC测定。色谱柱为Agilent C18柱(250mm×4.6mm,5μm),流动相为40mmol·L-1醋酸铵水溶液-乙腈(55∶45),流速1.0mL·min-1,检测波长230nm。结果:多西紫杉醇线性范围为15~960μg·L-1,方法回收率在98.4%~103.4%之间,提取回收率为72.3%~78.9%,日内和日间的精密度均小于9.7%。结论:该方法样品处理过程简便,重复性好,符合生物样品的测定要求,并适用于多西紫杉醇血药浓度的测定。
Objective: To establish a method for the determination of docetaxel in human plasma by high performance liquid chromatography (HPLC). Methods: 500μL of plasma sample was taken as the internal standard. With 1mol·L-1 sodium bicarbonate alkalized and extracted with diethyl ether, the supernatant was evaporated to dryness and dissolved in 150μL of mobile phase. 20μL was used for HPLC. The column was an Agilent C18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of 40 mmol·L -1 ammonium acetate in water (55:45) at a flow rate of 1.0 mL · min -1 and a detection wavelength of 230 nm. Results: The linear range of docetaxel was 15 ~ 960μg · L-1. The recoveries were between 98.4% and 103.4%. The recoveries were between 72.3% and 78.9%. The intra- and inter-day precision was less than 9.7%. Conclusion: The method has the advantages of simple sample processing, good repeatability, in line with the determination requirements of biological samples, and is suitable for the determination of docetaxel blood concentration.
