目前干扰素(IFN_s)是最有前途的治疗慢性乙型肝炎病毒(HBV)感染的抗病毒制剂。本文用HBV DNA转染的肝胚细胞瘤细胞株(HepG2.2.15)研究了IFNs对HBV复制和肝细胞基因表达的影响。作者将HepG2.2.15细胞培养在Dulbecco′s改良的Eagle′s培养基中(含10%胎牛血清),加入不同浓度(1×10~10~5IU/ml)的IFNs,每日换培养基和IFNs,共孵育5天。在除去IFNs后,逐日检测HBV的复制和IFNs介导的应答3天。结果:用~(32)P标记的HBV DNA探针进行Southern印迹检查时发现IFN-α2b和IFN-β均可暂时抑制HBV DNA复制。在与IFNs接触后的3天,
Currently interferon (IFN_s) is the most promising antiviral agent for the treatment of chronic Hepatitis B virus (HBV) infection. In this paper, HBV DNA transfected hepatoblastoma cell line (HepG2.2.15) investigated IFNs on HBV replication and hepatocyte gene expression. HepG2.2.15 cells were cultured in Dulbecco’s modified Eagle’s medium (containing 10% fetal bovine serum) and different concentrations (1 × 10 ~ 10 ~ 5IU / ml) of IFNs were added.
