目的:研究人TMJ滑膜细胞原代组织取材和体外培养方法,及其体外生长形态。方法:利用出生后死亡的胎儿进行TMJ滑膜组织取材,体外培养方法采用组织块法和关节腔内酶消化法,为观察滑膜细胞与其它细胞的区别,在同一胎儿个体上同时切取牙乳头和口腔粘膜组织各一块进行体外培养。结果:滑膜、牙乳头和口腔粘膜组织块原代培养后第3天出现活细胞生长,7d~10d分别传出第一代,传代培养生长稳定,每3天可传一代,分别传到38~40代,体外生长细胞形态有差异,滑膜细胞呈分散、网格状排列,其它细胞呈方向性排列。结论:TMJ滑膜细胞可采用组织块法进行体外培养,该细胞有自身的生长特点,可用其进行TMJ相关问题的研究
OBJECTIVE: To study the methods of primary culture of human TMJ synovial cells and their in vitro culture methods and their in vitro growth patterns. Methods: The fetuses born after death were harvested from TMJ synovial tissue. Tissue explant method and intra-articular enzyme digestion method were used to culture in vitro. To observe the difference between synovial cells and other cells, the dental papilla was cut simultaneously on the same fetus And oral mucosa of each one for in vitro culture. RESULTS: The live cells grew on the 3rd day after primary culture in synovium, dental papilla and oral mucosa tissue. The first generation came out on the 7th day to the 10th day respectively. The subculture culture grew steadily and passed every three days, reaching 38 ~ 40 passages, there were differences in the morphology of in vitro growth cells, synovial cells were scattered, grid-like arrangement, the other cells were arranged in a directional manner. CONCLUSION: TMJ synoviocytes can be cultured in vitro by tissue block method. The cells have their own growth characteristics and can be used for TMJ related research
