目的 :观察健脾补肾方药对实验性脾虚证大鼠环核苷酸及甲状腺激素水平的影响 ,探讨该方防治脾虚证的作用机理及脾与肾相互关系的意义。方法 :选用SD雄性大鼠 ,随机分为 4组 :即正常对照组 ,脾虚模型组 ,健脾补肾方高、低剂量组 ,采用大黄复制脾虚证动物模型 ,观察各组动物血浆环磷酸腺苷 (cAMP)、环磷酸鸟苷 (cGMP)及血清甲状腺素 (T4)、三碘甲腺原氨酸 (T3 )、促甲状腺素 (TSH)水平的变化。结果 :脾虚模型组大鼠血浆cAMP含量及cAMP/cGMP比值均比正常对照组升高 (p <0 0 1) ,T3 、T4含量比正常对照组下降 (p<0 0 1)。而健脾补肾方药能降低实验性脾虚证大鼠体内cAMP含量及cAMP/cGMP比值 (p <0 0 1) ,提高T3 、T4含量 (p <0 0 1)及脾脏、胸腺比值 ,又可使肾脏组织重量减轻等作用。 结论 :健脾补肾中药可以消除大黄的药物影响 ,防止出现脾虚症候 ,其作用机理与调节cAMP、cGMP及甲状腺激素水平有关。脾肾相关理论对临床有指导意义
Objective: To observe the effect of Jianpi Bushen Prescription on cyclic nucleotide and thyroid hormone levels in rats with experimental spleen deficiency syndrome, and to explore the mechanism of action of spleen-deficiency syndrome and the significance of the relationship between spleen and kidney. METHODS: SD male rats were randomly divided into 4 groups: normal control group, spleen-deficiency model group, and high-dose and low-dose groups of Jianpi Bushenfang. The spleen-deficiency syndrome animal model was made by using rhubarb to observe the plasma cAMP level in each group. Changes in cAMP, cGMP, and serum thyroxine (T4), triiodothyronine (T3), and thyrotrophin (TSH) levels. RESULTS: Compared with the normal control group, the plasma cAMP and cAMP/cGMP ratios in the spleen deficiency model group were higher than those in the normal control group (p < 0 01). The contents of T3 and T4 were lower than those in the normal control group (p <0 01). The Jianpi Bushen Prescription can reduce the cAMP content and cAMP/cGMP ratio (p < 0 01), increase the T3, T4 content (p <0.01) and spleen, thymus ratio in experimental spleen deficiency syndrome rats. Conclusion : The traditional Chinese medicine for invigorating the spleen and tonifying the kidney can eliminate the influence of rhubarb and prevent the appearance of symptoms of spleen deficiency.
