目的对2007年5月底,在北京市大兴区发生的一起手足口病(Hand Foot Mouth Disease,HFMD)疫情进行实验室分析鉴定。方法采集HFMD患儿的疱疹液、咽拭子、粪便标本,采用逆转录-聚合酶链反应(Reverse Tran-scription-Polymerase Chain Reaction,RT-PCR)扩增标本及病毒阳性分离物中人肠道病毒(Human Enterovirus,HEV)特异性核酸片段,比较RT-PCR方法和HEV分离的敏感性。然后利用测序方法对HEV阳性分离物进行HEV分型鉴定,确定本次疫情中HEV的型别。结果5例HFMD患儿的10份标本中,有8份测到HEV特异核酸片段,检出阳性率为80%,高于HEV分离阳性率(30%)。5例患儿和9例密切接触者的共19份标本中共分离到5株HEV,其中来自2例患儿和1例密切接触者的4株病毒为EV71型(HEV71),来自密切接触者的另外1株HEV为柯萨奇病毒A组16型。结论北京市大兴区5月底发生的一起HFMD疫情是由HEV71型引起的。
Objective To carry out laboratory analysis and identification of an outbreak of Hand Foot Mouth Disease (HFMD) in Daxing District of Beijing in late May 2007. Methods Herpes fluid, throat swabs and stool samples were collected from children with HFMD. The samples were collected and reverse transcriptase-polymerase chain reaction (RT-PCR) Virus (Human Enterovirus, HEV) -specific nucleic acid fragments, compared RT-PCR method and HEV separation sensitivity. HEV typing was then performed on HEV positive isolates by sequencing to determine the type of HEV in this outbreak. Results Of the 10 samples of 5 HFMD cases, 8 of them detected HEV specific nucleic acid fragments, and the positive rate was 80%, higher than the positive rate of HEV isolation (30%). Five HEVs were isolated from 19 samples of 5 children and 9 close contacts, of which 4 were EV71 (HEV71) from 2 children and 1 close contact, and those from close contacts Another HEV was coxsackievirus A group 16. Conclusion The HFMD outbreak in Daxing district of Beijing at the end of May was caused by HEV71 type.
