本文报告了生物素探针(Bio-HBV)试剂盒的临床应用研究。结果表明最低可检出0.1pgHBVDNA。Bio-HBV仅与HBVDNA杂交,不与PBR_(322)、λ、CT、HSDNA杂交。80例HBsAg献血员中,64例DNA阳性(80.0%),72例HBeAg阳性中,63例DNA阳性(87.5%)。符合率检测结果,敏感性97.4%,特异性92.3%,假阳性7.7%,假阴性2.6%,符合率98.1%,阳性预测值97.4%。Southern Blot试验表明89-1消毒剂30秒钟可杀灭200ng克隆HBVDNA。结果提示Bio-HBV可代替~(32)P HBV。
This article reports the clinical application of the biotin probe (Bio-HBV) kit. The results showed that the lowest detectable 0.1pgHBVDNA. Bio-HBV hybridized only with HBVDNA and did not hybridize with PBR_ (322), λ, CT, HSDNA. Of the 80 HBsAg blood donors, 64 were DNA positive (80.0%) and 72 of 72 HBeAg positive were DNA positive (87.5%). Conformity rate test results, sensitivity 97.4%, specificity 92.3%, false positive 7.7%, false negative 2.6%, coincidence rate of 98.1%, a positive predictive value of 97.4%. The Southern Blot test showed that 89-1 disinfectant killed 200 ng of cloned HBVDNA for 30 seconds. The results suggest that Bio-HBV can replace ~ (32) P HBV.
