目的观察龟羚帕安丸对帕金森病(PD)大鼠中脑黑质IL-1β、IL-6、i NOS、IFN-γ表达的影响。方法采用6-OHDA注射法建立PD大鼠模型,造模成功大鼠随机分为模型组,美多芭组(10mg·kg-1),龟羚帕安丸高、中、低剂量组(4,2,1g·kg-1),同时设立假手术组。假手术组、模型组给予等容积生理盐水,其他组予相应药物,连续灌胃给药30天。免疫组织化学法检测中脑黑质IL-1β、IL-6、i NOS、IFN-γ蛋白表达。结果龟羚帕安丸能明显抑制炎症因子IL-1β、IL-6、i NOS、IFN-γ表达。结论龟羚帕安丸通过降低IL-1β、IL-6、i NOS、IFN-γ表达,通过抗炎作用,对多巴胺能神经元产生保护作用,可能是龟羚帕安丸治疗PD的作用机制之一。
Objective To observe the effect of Guiling Pan’an pill on the expression of IL-1β, IL-6, iNOS and IFN-γ in substantia nigra of Parkinson’s disease (PD) rats. Methods PD rats were established by 6-OHDA injection. The rats were randomly divided into model group, model group, metoprol group (10 mg · kg -1), high, medium and low dose groups 2,1 g · kg-1), while setting up sham operation group. Sham operation group, model group were given equal volume of normal saline, other groups were given the corresponding drugs, continuous oral administration of 30 days. Immunohistochemistry was used to detect the expression of IL-1β, IL-6, iNOS and IFN-γ in midbrain substantia nigra. Results Guipan Pa’an pill can significantly inhibit the expression of IL-1β, IL-6, iNOS and IFN-γ. Conclusion Guinling Pu’an pill can protect dopaminergic neurons through decreasing the expression of IL-1β, IL-6, i NOS and IFN-γ, and may be one of the mechanisms of the therapeutic effect of Guilingpa’an pill on PD .
