在兔和小鼠胚胎发生的早期阶段,已注意到存在一种人绒毛膜促性腺激素(HCG)样物质。但有些研究使用生物学测定技术未能证实这一发现。本研究试图确定兔的桑椹胚是否含有一种用免疫荧光和免疫组织化学技术能够检出的CG样的物质。在雌兔与证明有生育力的雄兔交配后48小时处死雌兔,切下生殖道,用台氏液冲洗输卵管,从冲洗液中回收到39个囊胚。用磷酸盐缓冲液冲洗,保存于台氏液中的桑椹胚后将25个桑椹胚转移至含有兔抗HCGβ亚基(第一抗体)的凹面载玻片上孵育
In the early stages of rabbit and mouse embryogenesis, it has been noted that there is a human chorionic gonadotropin (HCG) -like substance. However, some studies using biological assay techniques failed to confirm this finding. This study sought to determine if a rabbit’s morula contains a CG-like substance that can be detected by immunofluorescence and immunohistochemistry. The rabbits were sacrificed 48 hours after the mating of the female rabbit with the male fertilized. The reproductive tract was excised and the tubal was washed with Tyrode’s solution. 39 blastocysts were recovered from the washings. After rinsing with Phosphate buffer and preserving morula in Tyrode’s solution, 25 morulae were transferred to a concave slide containing rabbit anti-HCG beta subunit (primary antibody)
