目的 :观察密蒙花对血管内皮细胞增生的抑制作用。方法 :制备含中药密蒙花的大鼠血清 ,将其加入到培养的内皮细胞中 ,应用流式细胞仪观察细胞增生与凋亡的情况。结果 :含密蒙花的大鼠血清对血管内皮细胞周期有明显影响 ,可使生长因子诱导的人脐静脉内皮细胞 (HUVEC)周期中G2 M期细胞比例减少 ,S期细胞比例相对增加 ,说明密蒙花可阻滞经VEGF刺激的HUVEC由S期进入G2 M期 ,从而降低其有丝分裂能力。密蒙花可诱导HUVEC凋亡。结论 :密蒙花对HUVEC的增生有抑制作用 ,从而为临床治疗视网膜血管增生性病变提供实验理论依据。
Objective : To observe the inhibitory effect of Buddleja officinalis on proliferation of vascular endothelial cells. METHODS: Rat serum containing Buddlejae was prepared and added to cultured endothelial cells. Cell proliferation and apoptosis were observed by flow cytometry. RESULTS: The serum of rats with Buddleja officinalis had a significant effect on the cell cycle of vascular endothelial cells, and the ratio of G2 M phase cells in the human umbilical vein endothelial cells (HUVEC) induced by growth factors was reduced, and the proportion of cells in S phase was relatively increased. Buddleia flower can block HUVEC stimulated by VEGF from S phase to G2 M phase, thereby reducing its mitogenic ability. Buddleia can induce HUVEC apoptosis. Conclusion: Buddleja officinalis inhibited the proliferation of HUVEC, and provided theoretical basis for clinical treatment of retinal vascular proliferative lesions.
