建立一种同时测定绿原酸、咖啡酸、3-羟基苯丙酸(3-hydroxyphenylpropionic acid,3-HPA)、p-香豆酸(p-coumaric acid,PA)、阿魏酸(ferulic acid,FA)和马尿酸(hippuric acid,HA)含量的高效液相色谱方法。色谱柱为美国Agilent EclipseXDB-C18(250mm×4.6mm,5μm);流动相为甲醇和1%的冰醋酸水溶液,梯度洗脱;检测波长为320nm和253nm;流速为1mL/min。六种酚酸物质在一定范围内有良好的线性关系,相关系数为0.9979~0.9999,平均回收率为86.84%~101.56%,相对标准偏差为1.15%~2.29%。结果表明该方法快速、准确、灵敏度高,可用于绿原酸及其代谢物的检测。
To establish a method for the simultaneous determination of chlorogenic acid, caffeic acid, 3-hydroxyphenylpropionic acid (3-HPA), p-coumaric acid (PA), ferulic acid FA) and hippuric acid (hippuric acid, HA) content of high performance liquid chromatography method. The column was Agilent EclipseXDB-C18 (250mm × 4.6mm, 5μm). The mobile phase consisted of methanol and 1% aqueous glacial acetic acid with gradient elution. The detection wavelength was 320nm and 253nm. The flow rate was 1mL / min. The six phenolic acids have a good linear relationship in a certain range, the correlation coefficient is 0.9979 ~ 0.9999, the average recovery rate is 86.84% ~ 101.56%, the relative standard deviation is 1.15% ~ 2.29%. The results show that the method is rapid, accurate and sensitive and can be used for the detection of chlorogenic acid and its metabolites.
