目的:通过对比MDA- NeoR 与MDA 人乳癌细胞的生物学特征,阐明原核标志基因NeoR 的长期稳定整合与表达是否对乳癌细胞原有的生物学特性产生影响。方法:对已建立且证实表达NeoR 基因> 6 个月的7 株MDA-NeoR 细胞系进行G418 抗性、表面抗原、细胞倍增时间、细胞周期、肿瘤细胞集落形成及裸鼠体内成瘤能力的检测。结果:MDA- NeoR 细胞对高浓度的G418 仍产生抗性并表达Neo R 基因;其细胞倍增时间、S 期分数、肿瘤细胞体外集落形成率与MDA 无明显区别( P> 0-05) ;角蛋白CK19 与表面膜抗原EMA 的表达在两组是等同的;并且MDA- NeoR 7 细胞系在裸鼠体内的成瘤能力与MDA 细胞也是相似的。结论:外源标志基因NeoR 的长期稳定整合与表达对MDA 人乳癌细胞的原有生物学特性无明显影响
Objective: To compare the biological characteristics of MDA-NeoR and MDA human breast cancer cells to elucidate whether the long-term stable integration and expression of prokaryotic marker gene NeoR affects the original biological characteristics of breast cancer cells. METHODS: G418 resistance, surface antigen, cell doubling time, cell cycle, tumor cell colony formation, and tumorigenicity in nude mice were determined in 7 MDA-NeoR cell lines that had been established and confirmed to express NeoR genes for > 6 months. . RESULTS: MDA-NeoR cells were still resistant to high concentrations of G418 and expressed the Neo R gene; their cell doubling time, S-phase fraction, and tumor cell formation in vitro were not significantly different from those of MDA (P> 0-05); The expression of protein CK19 and surface membrane antigen EMA is equivalent in both groups; and the tumorigenicity of MDA-NeoR7 cell line in nude mice is similar to that of MDA cells. Conclusion: The long-term stable integration and expression of exogenous marker gene NeoR has no significant effect on the original biological characteristics of MDA human breast cancer cells.
