本研究旨在探讨白藜芦醇甙(polydatin)对大鼠缺血/再灌注心肌细胞凋亡的影响及其相关机制。成年雄性Sprague-Dawley大鼠随机分为三组:对照(CON)组,缺血/再灌注(ischemia/reperfusion,I/R)组,白藜芦醇甙(POLY,50μmol/L)组。应用Langendorff灌注技术,给予心脏30min缺血/60min再灌注,通过TUNEL和流式细胞技术检测细胞凋亡以及Bcl-2和Bax蛋白的表达。结果显示:(1)与CON组相比,I/R组心肌缺血/再灌注后心肌凋亡细胞(TUNEL染色)数量、凋亡率明显增加;(2)与I/R组相比,POLY组心肌缺血/再灌注后心肌凋亡细胞(TUNEL染色)数量明显减少[(18.1±4.0)%vs(35.1±5.4)%,P<0.01];(3)流式细胞术测定显示I/R组心肌细胞凋亡率为(15.43±4.55)%,明显高于POLY组心肌[(8.66±3.18)%](P<0.01);(4)I/R组心肌Bax蛋白表达明显高于POLY组心肌(P<0.05),POLY组心肌Bcl-2蛋白表达及Bcl-2/Bax比值明显高于I/R组心肌(P<0.05,P<0.01)。以上结果表明,白藜芦醇甙可通过增加Bcl-2蛋白表达,减少Bax蛋白表达而减少I/R诱导的大鼠心肌细胞凋亡。
This study was to investigate the effect of polydatin on cardiomyocyte apoptosis induced by ischemia/reperfusion in rats and its related mechanism. Adult male Sprague-Dawley rats were randomly divided into three groups: control (CON) group, ischemic/reperfusion (I/R) group, and resveratrol (POLY, 50 μmol/L) group. Langendorff perfusion technique was used to give 30 min ischemia/60 min reperfusion in the heart. Cell apoptosis and Bcl-2 and Bax protein expression were detected by TUNEL and flow cytometry. The results showed that: (1) Compared with CON group, the number and apoptosis rate of apoptotic cells (TUNEL staining) increased significantly in I/R group after myocardial ischemia/reperfusion; (2) Compared with I/R group, The number of apoptotic myocytes (TUNEL staining) was significantly decreased in the POLY group after myocardial ischemia/reperfusion ((18.1±4.0)% vs (35.1±5.4)%, P<0.01); (3) Flow cytometry showed I The apoptosis rate of cardiomyocytes in /R group was (15.43±4.55)%, which was significantly higher than that of POLY group [(8.66±3.18)%] (P<0.01); (4) The expression of Bax protein was significantly higher in I/R group. In the POLY group, the expression of Bcl-2 protein and the ratio of Bcl-2/Bax were significantly higher in the POLY group than in the I/R group (P<0.05, P<0.01). The above results show that resveratrol can reduce I/R-induced cardiomyocyte apoptosis in rats by increasing Bcl-2 protein expression and reducing Bax protein expression.
