目的:探讨Livin基因在人胆管癌组织及胆管癌细胞系中的表达情况及其与胆管癌发生发展之间的关系。方法:采用免疫组织化学技术(SP法)检测Livin基因蛋白在45例人胆管癌标本及及40例癌旁胆管组织、20例正常胆管组织标本中的表达;同时采用RT-PCR法及SP法检测了Livin基因mRNA和蛋白在人胆管癌细胞系QBC939及非肿瘤细胞系HT-1080中表达。结果:Livin在胆管癌组织中表达阳性率为57.8%,而癌旁胆管组织、非癌胆管组织中未能检测到Livin表达。Livin表达与性别、年龄、肿瘤大小及肿瘤分化程度无关。在有淋巴结转组中,Livin阳性表达率(70.4%)明显高于无淋巴结转移组(38.9%)。在人胆管癌细胞QBC939中,Livin mRNA及蛋白均特异性表达,而非肿瘤细胞系HT-1080未见Livin表达。结论:Livin基因在人类胆管癌组织和细胞系中选择性高表达,其可能与胆管癌发生、发展及预后密切相关。
Objective: To investigate the expression of Livin gene in human cholangiocarcinoma and cholangiocarcinoma cell lines and its relationship with the occurrence and development of cholangiocarcinoma. Methods: The expression of Livin gene protein was detected by immunohistochemistry (SP method) in 45 specimens of human cholangiocarcinoma and 40 specimens of para-cancerous bile duct tissue and 20 specimens of normal bile duct tissue. RT-PCR and SP The expression of Livin mRNA and protein was detected in human cholangiocarcinoma cell line QBC939 and non-tumor cell line HT-1080. Results: The positive expression rate of Livin in cholangiocarcinoma was 57.8%, while the expression of Livin was not detected in the paracancerous bile duct tissue and non-carcinoma cholangiocarcinoma. Livin expression was not related to sex, age, tumor size and tumor differentiation. In lymph node metastasis group, the positive expression rate of Livin (70.4%) was significantly higher than that of the group without lymph node metastasis (38.9%). In human cholangiocarcinoma cells QBC939, Livin mRNA and protein were all specifically expressed, while no Livin expression was found in non-tumor cell line HT-1080. Conclusion: Livin gene is selectively expressed in human cholangiocarcinoma tissues and cell lines, which may be closely related to the occurrence, development and prognosis of cholangiocarcinoma.
