珠子参水提物预处理对小鼠脑缺血再灌注损伤的影响

目的研究珠子参水提物预处理对小鼠局灶性脑缺血再灌注损伤的保护作用。方法雄性5周龄昆明小鼠170只,采用完全随机设计分组法分为6组:假手术组(n=20)、模型组(n=30)、尼莫地平组(2 mg/kg,n=30)和珠子参水提物低、中、高剂量组(2.5、5.0、10.0 g/kg,n=30)。除假手术组和模型组灌胃给予蒸馏水20 ml/kg外,其余各组灌胃给予相应剂量的药物。以上各组给药7 d后制作大鼠大脑中动脉闭塞模型,假手术组切开缝合不进行动脉阻塞,在缺血2 h再灌注24 h后进行行为学检测,记录各组小鼠存活率。取大脑并用2,3,5-三苯基氯化四氮唑染色后测定梗死面积和脑含水量;制作脑匀浆测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)、黄嘌呤氧化酶(XOD)酶活性和丙二醛(MDA)含量。结果珠子参水提物中、高剂量组的成活率分别为77.8%(21/27)和82.1%(23/28),与模型组[51.9%(14/27)]比较具有统计学差异(P<0.05);珠子参水提物高剂量组的神经症状评分(1.77±0.38)、斜板角度(63.6±5.5)°优于模型组[神经症状评分(2.21±0.43),斜板角度(55.1±7.5)°](P<0.05,P<0.01),与尼莫地平效果接近;与模型组比较,珠子参水提物中、高剂量组能显著降低脑梗死面积和脑含水量(P<0.05,P<0.01),并能明显增加SOD、GSH-PX、CAT酶活性(P<0.01),降低XOD酶活性和MDA含量(P<0.01);高剂量组在改善GSH-PX、SOD和CAT酶及XOD酶活性方面优于尼莫地平组。结论珠子参水提物预处理对小鼠局灶性脑缺血再灌注损伤具有明显的保护作用。 Objective To study the protective effect of aqueous extract of Radix Pilosulas against focal cerebral ischemia-reperfusion injury in mice. Methods A total of 170 Kunming mice, 5 weeks old, were randomly divided into 6 groups: sham operation group (n = 20), model group (n = 30), nimodipine group = 30), and low, medium and high doses of aqueous extract (2.5, 5.0, 10.0 g / kg, n = 30). In addition to the sham operation group and model group, intragastric administration of distilled water 20 ml / kg, the remaining groups were given the appropriate dose of drug. The rats in the above groups were given 7-day-old model of middle cerebral artery occlusion (MCAO). The rabbits in the sham-operation group were sutured without occlusion. The rats were sacrificed at 2 hours after ischemia and reperfusion for 24 hours. The survival rate . The brain was taken and infarct size and brain water content were measured after staining with 2,3,5-triphenyltetrazolium chloride. Brain homogenate was prepared to measure superoxide dismutase (SOD) and glutathione peroxidase GSH-PX), catalase (CAT), xanthine oxidase (XOD) activity and malondialdehyde (MDA) content. Results The survival rates of middle and high dose group were 77.8% (21/27) and 82.1% (23/28), respectively, which were significantly different from the model group (51.9%, 14/27) (1.77 ± 0.38), oblique angle (63.6 ± 5.5) ° higher than the model group [neurotic symptom score (2.21 ± 0.43), slope angle ( 55.1 ± 7.5) °] (P <0.05, P <0.01), which was similar to nimodipine.Compared with the model group, the medium and high dose of water extract of Beauveria bassiana could significantly reduce the infarct size and brain water content 0.05, P <0.01), and significantly increased the activities of SOD, GSH-PX and CAT (P <0.01) and decreased the activities of XOD and MDA (P <0.01) And CAT enzyme and XOD enzyme activity than nimodipine group. Conclusion Pretreatment with aqueous extract of Radix Phellodendron has a significant protective effect on focal cerebral ischemia-reperfusion injury in mice.
