目的研究葛根芩连配方颗粒(简称葛根芩连颗粒)在大肠湿热证模型大鼠尿液中的代谢产物。方法采用综合因素(高糖和高脂饮食+高温和高湿环境+肠产毒性大肠杆菌致病因子)制备大肠湿热证大鼠模型;利用HPLC-DAD-MS技术对大鼠ig葛根芩连颗粒后尿液中代谢产物进行分析,检测波长272 nm;流动相A为0.1%甲酸水溶液,流动相B为乙腈-甲醇(6∶4),梯度洗脱。结果从给予葛根芩连颗粒的大鼠尿液中发现并鉴定了5种主要代谢产物,分别为葛根素、异甘草素、甘草素、汉黄芩素、千层纸素A,其中葛根素来自葛根,汉黄芩素与千层纸素A来自黄芩,异甘草素与甘草素来自甘草。结论在大肠湿热证模型大鼠尿液中,葛根芩连颗粒中黄芩与甘草的代谢产物主要以苷元为主,葛根中的葛根素主要以原形排出。
Objective To study the metabolites of Gegen Qinlian formula particles (Gegen Qinlian particles) in the urine of rats with damp-heat syndrome of the large intestine. Methods The rat model of damp-heat syndrome of large intestine was established by using comprehensive factors (high-sugar and high-fat diet + high temperature and high humidity environment + pathogenic factor of enterotoxigenic Escherichia coli); HPLC-DAD- After the urine metabolites were analyzed, detection wavelength 272 nm; mobile phase A 0.1% formic acid in water, mobile phase B acetonitrile - methanol (6: 4), gradient elution. Results Five major metabolites were identified and identified from the urine of rats given Gegen Qinlian granules, namely puerarin, isoliquiritigenin, liquiritigenin, wogonin and melatonin A, respectively. Among them, puerarin came from Pueraria , Wogonin and Melaleuca paper A from skullcap, isoliquiritigenin and licorice root from licorice. Conclusions The metabolites of Scutellaria baicalensis and Glycyrrhizae in Gegen Qinlian Granules are mainly aglycone in the urine of rat model of damp-heat syndrome of the large intestine. The puerarin in Pueraria lobata mainly excretes in the prototype form.
