热水抽提蒸汽爆破处理后的玉米秸秆,利用高效液相色谱仪,AminexHPX-87P离子色谱柱为分析柱,示差折光检测器分离分析所得水解液中的主要糖分:木糖、葡萄糖、纤维二糖、半乳糖、阿拉伯糖、甘露糖。通过比选各种样品前处理方法及色谱条件优化,选定了添加适量三氯乙酸做样品前处理,检测采用85℃柱温和超纯水作为流动相,水解液样品中各组分的线性相关系数均在0.9990~0.9999之间,回收率为86.3%~109.4%,精密度为1.14%~4.64%。在样品中糖损失最小的情况下,有效去除了色素、脂类、蛋白等影响测定并损害色谱柱的污染物,在最佳色谱条件下,蒸汽爆破水解液中的主要糖分能够同时被测定。
