An HPLC method was developed for the determination of strychnine and brucin contained in SHEN JIN HUO LUO Pill. The analysis was carried out by using a spherisob cyano column (Φ 5 μm, 4.6×150 mm). The mobile phase consists of methanol water acetic acid triethylamin (9400∶465∶90∶45, v/v). Detection wavelenth was set at 254 nm, and calibration graphs were rectilinear over the concentration ranges of 4~20 μg/ml for strychnine. The regression equation was %A=-18020+11991C(r%=0.9999). Calibration graphs were rectilinear over the concentration ranges of 2~10 μg/ml for brucine. The regression equation was %A=-17356+6871C(r%=0.9974). The precision of sample determination was RSD=0.85% for strychine and 6.2% for brucine. The average recovery rate was 99.73% for strychine and 99.33% for brucine.
