目的:探讨回药爱康方(HAKF)含药血清对人肺腺癌A549细胞凋亡的影响。方法:SD大鼠随机分为对照组、爱康方低、中、高剂量组、环磷酰胺(CTX)组、回药爱康方低、中、高剂量分别联合环磷酰胺化疗组,药物低、中、高剂量15.0,30.0,45.0 g·kg-1,连续灌胃15 d,麻醉后心脏取血,制备含药血清。培养细胞,将细胞分为实验组和对照组,实验组分别给予含5%,10%,20%3个体积分数的相应各组含药血清,对照组给予等体积分数的对照组大鼠血清,培养24,48,72 h后采用Annexin-VFITC/PI双染法测定各组含药血清对A549细胞凋亡的影响。结果:与对照组比较,各时段5%,10%,20%浓度含药血清可提高A549的凋亡率(P<0.05),72 h时,各浓度回药爱康方低、中、高剂量含药血清对细胞的凋亡率的提高程度小于CTX(P<0.05);72 h 10%体积分数回药爱康方低剂量联合环磷酰胺血清提高细胞的凋亡率大于单纯CTX(P<0.05);各浓度爱康方中剂量和高剂量血清组,72 h与24 h比较,两组A549细胞的凋亡率均升高(P<0.05);随着血清体积分数的提高,各组细胞的凋亡率和坏死率呈增大趋势。结论:回药爱康方可诱导人肺腺癌A549细胞的凋亡,且有一定的剂量和时间依赖性。
Objective: To investigate the effect of HAKF-containing serum on the apoptosis of human lung adenocarcinoma A549 cells. At 72 hours, the drug concentration of low, medium and high Apoptosis rate of the drug-containing serum was lower than that of CTX (P <0.05); 72 h 10% volume fraction of Aikangfang low dose combined with cyclophosphamide serum increased the apoptosis rate of cells than CTX <0.05). The apoptosis rates of A549 cells in both groups were significantly increased at 72 h and 24 h (P <0.05) with the middle dose and high dose of Aikangfang, respectively. With the increase of serum volume fraction, The apoptosis rate and necrosis rate of the group of cells showed an increasing trend. Conclusion: Aikang Formula can induce the apoptosis of human lung adenocarcinoma A549 cells in a dose and time dependent manner.
