目的观察经典眨眼条件反射建立后豚鼠小脑齿状-中位核(D-Inuclei)突触超微结构的变化,探讨联合型学习和突触超微结构变化的关系。方法实验动物分为配对组(n=4)和非配对组(n=4)。采用500ms正弦波纯音和100ms束状氧气流进行配对刺激训练配对组动物,而非配对组动物则接受声音和气流的非配对刺激。训练6d后,电镜下观察各组动物小脑齿状-中位核突触超微结构,应用图像分析软件比较突触超微结构相关参数的差异。结果配对组动物在6d的条件反射训练后均可获得90%以上的条件反应率,而非配对组动物6d内条件反应率均低于10%;相对于非配对组,配对组动物小脑齿状-中位核突触密度无显著增加(P>0.05),但突触后膜致密区厚度增厚(P<0.05),突触活性区域增多(P<0.05)。结论经典眨眼条件反射建立可能与小脑齿状-中位核突触超微结构的变化有关。
Objective To observe the changes of synaptic ultrastructure in the cerebellar dentate nucleus (D-Inuclei) of the guinea pig after the establishment of classic wink reflex, and to explore the relationship between synaptic ultrastructure and synaptic ultrastructure changes. Methods The experimental animals were divided into paired group (n = 4) and unpaired group (n = 4). Matched stimuli were used to train pairing animals with 500 ms sine wave pure tone and 100 ms beam oxygen flow, whereas non-paired animals received unpaired stimulation of sound and airflow. After 6 days of training, the ultrastructure of dentate and medial nucleus synapse in each group were observed under electron microscope. The differences of synaptic ultrastructure parameters were compared by image analysis software. Results The response rate of conditioned animals in the paired animals was over 90% after 6 days of conditioned reflex training, while the response rate of all animals in the paired animals was less than 10% within 6 days. Compared with the non-paired animals, - There was no significant increase in the median synaptic density (P> 0.05), but the thickness of the synaptic membrane was thicker (P <0.05) and the area of synaptic activity increased (P <0.05). Conclusions The establishment of classic wink reflex may be related to the changes of the ultrastructure of dentate and medial synapses in cerebellum.
