目的观察自发性高血压大鼠(SHR)下丘脑视上核、室旁核内血管紧张素Ⅱ(AngⅡ)的变化。方法选择6,10,16周龄SHR和WKY大鼠各8只,采用AngⅡ单克隆抗体免疫组化S-P方法染色,计算机图像分析仪测定下丘脑视上核、室旁核内AngⅡ免疫反应阳性物质的染色程度。结果6,10,16周龄SHR血压逐渐升高,且显著高于同龄WKY大鼠。AngⅡ免疫反应阳性物质位于下丘脑视上核、室旁核大细胞部神经细胞的胞浆内。3个周龄组的SHR下丘脑视上核、室旁核内AngⅡ免疫反应阳性物质的染色强度均显著高于同龄的WKY大鼠。结论SHR下丘脑视上核、室旁核内AngⅡ的变化可能与其高血压的形成有关
Objective To observe the changes of angiotensin Ⅱ (Ang Ⅱ) in hypothalamic supraoptic nucleus and paraventricular nucleus in spontaneously hypertensive rats (SHR). Methods Eight SHR and WKY rats at 6, 10, and 16 weeks of age were selected and stained with Ang Ⅱ monoclonal antibody (S-P) by immunohistochemistry. The Ang Ⅱ immunoreactivity in supraoptic nucleus and paraventricular nucleus of hypothalamus was measured by computer image analyzer The degree of staining of positive substances. Results 6, 10, 16 weeks of age SHR blood pressure gradually increased, and significantly higher than WKY rats of the same age. Ang Ⅱ immunoreactive positive substance is located in the hypothalamic supraoptic nucleus, paraventricular nucleus of large cells in the cytoplasm of neurons. The staining intensity of Ang Ⅱ immunoreactive substance in SHR hypothalamic supraoptic nucleus and paraventricular nucleus in 3 weeks old group was significantly higher than that in WKY rats of the same age. Conclusion The changes of Ang Ⅱ in supraoptic nucleus and paraventricular nucleus of SHR may be related to the formation of hypertension
