骨巨细胞瘤(giant cell tumor of bone,GCT)是一种以多核巨细胞和单核基质细胞组成的常见骨肿瘤,在GCT中,多核巨细胞的存在,始终是该肿瘤的典型组织学特征之一.尽管在GCT的众多研究中发现,GCT中的多核巨细胞可能就是肿瘤环境中的破骨细胞[1~3],并可直接引起骨质的溶解[4,5].但一直未能找到一种有效的方法从GCT中提取高纯度的多核巨细胞,以供体外实验研究.1997年我们在完成国家教委博士点基金和霍英东教育基金课题“骨巨细胞瘤病变部位局部骨质吸收机制”的研究中[6,7],先后尝试运用了percoll密度梯离心法和50μm孔经不锈钢网过滤法,从GCT组织中提取多核巨细胞.结果证实,不锈钢网过滤法确为一种简便、经济而高效的分离提纯方法,值得介绍和推广.
