目的:探讨HOXA11基因在卵巢颗粒细胞内的异常表达与PCOS的相关性。方法:采用免疫荧光法、RT-PCR法、Western-blot法检测颗粒细胞中HOXA11的表达水平。结果:①在PCOS组中,镜下细胞内见明显的红色荧光主要定位于细胞核,细胞液中可见少许微弱红色荧光;对照组中,细胞核和细胞液中可见更强的红色荧光。PCOS组与对照组比较,细胞核和细胞液中HOXA11的表达都明显减少(P<0.05)。②PCOS组HOXA11 mRNA的表达水平较对照组低(P<0.05)。③Western-blot结果显示,PCOS组与对照组的颗粒细胞中均有HOXA11蛋白表达。PCOS组表达较弱,对照组表达较强(P<0.05)。免疫荧光、RT-PCR、Western-blot均提示PCOS组颗粒细胞中HOXA11表达水平较对照组低(P<0.05)。结论:HOXA11在PCOS组的颗粒细胞中表达降低,提示其可能在PCOS发生发展过程中起着重要作用。
Objective: To investigate the correlation between the abnormal expression of HOXA11 gene in ovarian granulosa cells and PCOS. Methods: The expression of HOXA11 in granulosa cells was detected by immunofluorescence, RT-PCR and Western-blot. Results: ① In the PCOS group, obvious red fluorescence was found in the nucleus under the microscope and a little weak red fluorescence was observed in the cell liquid. In the control group, more red fluorescence was observed in the nucleus and cytosol. Compared with the control group, the expression of HOXA11 in nucleus and cytosol of PCOS group was significantly decreased (P <0.05). ② The expression of HOXA11 mRNA in PCOS group was lower than that in control group (P <0.05). Western-blot results showed that HOXA11 protein was expressed in both granulosa cells of PCOS group and control group. The expression of PCOS group was weaker and the expression of the control group was stronger (P <0.05). Immunofluorescence, RT-PCR and Western-blot indicated that HOXA11 expression in PCOS group was lower than that in control group (P <0.05). Conclusion: The decreased expression of HOXA11 in PCOS group of granulosa cells suggests that it may play an important role in the development of PCOS.
