目的探讨伤科接骨片在下颌骨缺损修复中的作用机制。方法健康雄性新西兰兔72只,随机分为正常对照组(24只)、模型组(24只)和伤科接骨片组(24只),建立下颌骨缺损模型。正常对照组、模型组动物给予正常饲料,伤科接骨片组动物给予伤科接骨片饲料。分别于建模后第7、14、28、56天处死相同例数动物(n=6),收集下颌骨缺损处骨组织块。采用RT-PCR技术检测骨保护素(osteoprotegerin,OPG)、骨保护素配体(osteoprotegerin ligand,OPGL)基因表达,免疫组化检测骨组织染色强度和面积。结果与正常对照组比较,模型组兔下颌骨组织OPGmRNA表达在建模后第7天显著上调(P<0.05),OPGL mRNA表达在建模后第14天显著上调(P<0.05);与模型组比较,伤科接骨片组建模后第14、28、56天OPGmRNA表达均上调明显(P<0.05),骨组织阳性染色更强,面积更广,建模后第14天OPGL mRNA表达下调明显(P<0.05),骨组织阳性染色更弱,面积更小,OPG mRNA/OPGL mRNA比值在建模后第14、28、56天明显上调(P<0.05)。结论伤科接骨片促进下颌骨缺损修复的作用可能与其调节OPGmRNA、OPGL mRNA表达水平有关。
Objective To investigate the mechanism of Shangke Jiegu tablets in the repair of mandibular defects. Methods 72 healthy male New Zealand rabbits were randomly divided into normal control group (24 rats), model group (24 rats) and Shangke Jiegu tablets group (24 rats) to establish a model of mandibular defect. The normal control group and model group animals were given normal feed, and the injuries were given to the animals of Shangke Jiegu Tablet group. The same number of animals (n = 6) were sacrificed on the 7th, 14th, 28th and 56th day after modeling, and the bone tissue pieces of the mandible defect were collected. The expression of osteoprotegerin (OPG) and osteoprotegerin ligand (OPGL) gene was detected by RT-PCR. The intensity and area of bone tissue were detected by immunohistochemistry. Results Compared with the normal control group, OPG mRNA expression in the mandibular tissue of model group was significantly increased on the 7th day after modeling (P <0.05), and the expression of OPGL mRNA significantly increased on the 14th day after modeling (P <0.05) OPG mRNA expression was significantly increased (P <0.05) on the 14th, 28th and 56th day after injury in the Shangke Jie Goup tablet group. OPG mRNA expression in the Shangkejiegu tablets group was stronger than the control group (P <0.05). The positive staining of bone tissue was weaker and the area was smaller. The ratio of OPG mRNA / OPGL mRNA increased significantly (P <0.05) on the 14th, 28th and 56th day after modeling. Conclusion The effect of Shangke Jiegu tablet in promoting the repair of mandibular defects may be related to the regulation of OPG mRNA and OPGL mRNA expression.
